Do i have to mix gas with oil on the first start up?

I just built a motorized bike completely perfect with no flaws on its parts i'm pretty sure. My dad however wants to test the bike out WITHOUT mixing the gas and oil to 16:1 ratio. Would the engine start up on the first try? it's a 2-Stroke engine.
